This is the first Coen brothers film where Joel Coen and Ethan Coen are both given directing and producing credits. They have shared these duties on all of their films, but Joel has always been listed as the director, and Ethan as producer.

Throughout the film, Mrs. Munson says that she donates monthly to Bob Jones University, citing it as a "good Bible college". Bob Jones University banned black students until the 1970s, and did not allow interracial dating until 2000.
When the group is debating various methods to kill Mrs. Munson, Professor Dorr suggests shackling her in an earthen recess in the wall and enclosing it with the leftover bricks and mortar they have available. This is a reference to "A Cask of Amontillado" by Edgar Allan Poe, whom Dorr cites frequently throughout the film.
The first piece of music that Dorr's band pretends to play is Boccherini's minuetto: it was the same piece of music that was played by Professor Marcus's band in the original The Ladykillers (1955).
Prior to filming, Tom Hanks had not seen The Ladykillers (1955) as he did not want it to prejudice the way he acted in the remake.

The exterior shot of the riverboat casino is an actual casino on the Mississippi River in Vicksburg, Mississippi. The Ameristar lettering on the "paddlewheel" was edited out and the movie casino's name put in its place. In the movie, the casino has closing hours while the Ameristar casino is a 24/7 operation (the entire parking lot beside the boat was emptied for the shot). Additionally, 2 large river bridges that would normally be at the left side of the frame are edited out also.
All the musical instruments were re-created by Danny Ferrington, guitar-maker craftsman, because, according to him, the owners of authentic antique instruments refused to lend them for the film. The strange "triple guitar" is a harp-lute - an unknown ancient instrument that nobody knows how to play and for which no music was ever written - that Ferrington had to build from scratch.
Several times at the beginning of the film, when Mrs. Munson is complaining about a neighbor's loud music, she repeats lyrics to the song to illustrate her displeasure. The song is "I Left My Wallet in El Segundo", by A Tribe Called Quest. It was released in 1990.

The portrait of Othar is based on Alec Guinness. The original movie both starred Guinness and had his likeness in the portrait, which itself is a reference to one of the eight characters Guinness played in Kind Hearts and Coronets.
In the movie, J.K. Simmons paraphrases the classic line from Spiderman, "With great power, comes great responsibility." Sam Raimi directed the big budget version, where Simmons played J Jonah Jameson, and Raimi is also a close friend of Joel and Ethan Coen, the directors of “The Ladykillers”.
